Bu çalışma, diyabet hastalığının akciğer dokusu üzerindeki etkilerini incelemek amacıyla ratlarda bir diyabet modeli kullanarak histopatolojik görüntülerin analiz ve sınıflandırılmasını hedeflemektedir. Çalışmanın başlangıcında, kontrol ve Streptozotosin (STZ) ile diyabetik gruplar oluşturulmuştur. Akciğer dokusundaki değişiklikleri incelemek için kaspaz immunohistokimyasal boyama kullanılmıştır. Görüntülerden Yerel İkili Örüntüler (Local Binary Pattern, LBP) ve Gri Seviye Eş Oluşum Matrisi (Gray-Level Co-Occurrence Matrix, GLCM) gibi özellikler elde edilmiştir. Bu özellikler ile diyabetin akciğer dokusuna etkilerini analiz etmek amacıyla histopatolojik görüntüler analiz edilmiştir . Daha sonra, Lasso yöntemi ile en önemli özellikler seçilmiş ve kullanılmıştır. Elde edilen özellikler, Destek Vektör Makinesi (Support Vector Machine, SVM), K-en Yakın Komşu (K-nearest neighbors, KNN), Yapay Sinir Ağları (YSA) ve Karar Ağacı (Decision Tree, DT) gibi dört farklı sınıflandırma yöntemi ile sınıflandırılmıştır. Bu yöntemler, görüntülerin sınıflandırılmasında kullanılmış ve görüntülerde sınıflandırma sonuçları elde edilmiştir. Kırmızı ve mavi kanallardan elde edilen görüntüler ile en iyi sınıflandırma performansı sırasıyla %91.08 ve %93.87 doğruluk oranlarıyla YSA sınıflandırıcısıyla elde edilirken, yeşil kanaldan elde edilen görüntüler ile en yüksek doğruluk oranı %87.15 olarak SVM sınıflandırıcısıyla elde edilmiştir. Bu sonuçlara göre, histopatolojik görüntü analizi yoluyla diyabetin akciğer dokularına etkisini objektif bir şekilde değerlendirmek için LBP, GLCM özellikleri ve makine öğrenme algoritmalarından oluşan sınıflandırma modelinin önemli bir potansiyele sahip olduğu görülmektedir.
This study aims to analyze and classify histopathological images using a rat model of diabetes to examine the effects of diabetes on lung tissue. At the beginning of the study, control and diabetic groups were established using Streptozotosin (STZ). Caspase immunohistochemical staining was used to examine changes in lung tissue. Features such as Local Binary Patterns (LBP) and Gray-Level Co-Occurrence Matrix (GLCM) were extracted from the images. These features were analyzed to assess the impact of diabetes on lung tissue. Subsequently, the most important features were selected and used with the Lasso method. The obtained features were classified using four different classification methods: Support Vector Machine (SVM), K-nearest neighbors (KNN), Artificial Neural Networks (ANN), and Decision Tree (DT). These methods were used for image classification, and classification results were obtained. The best classification performance was achieved with images obtained from the red and blue channels, with accuracy rates of 91.08% and 93.87% using the ANN classifier, while images obtained from the green channel yielded the highest accuracy rate of 87.15% with the SVM classifier. According to these results, it is evident that a classification model comprising LBP, GLCM features, and machine learning algorithms has significant potential for objectively assessing the impact of diabetes on lung tissues through histopathological image analysis.
